Resumo: dc.description.abstractThis study aims to assess the nuclear damage rate and the DNA fragmentation in erythrocytes from lambari Astyanax aff. paranae, by Piscine Micronucleus Test and Comet Assay. There were performed two fish collections, in winter 2011 and summer 2011, and in three sampled sites, two of these in Campo River (upstream and downstream of Campo Mourão city) and the third site was located in Perobas Biological Reserve (REBIO), in Tuneiras do Oeste city- Paraná State. In laboratory, the specimens were anaesthetized for blood collection by cardiac puncture. For Piscine Micronucleus Test, the blood samples were fixed in microscope lamina, stained with 10% Giemsa, and were computed 1000 erythrocytes. For Comet Assay, the blood samples were treated to remove the cell membrane, following the electrophoresis in alkaline conditions and staining with ethidium bromide. There were computed 100 cells, assessing the DNA fragmentation intensity. Through ANOVA, we can observe that the sample site in Biological Reserve during summer presents the highest nuclear alteration rate and the sites upstream in summer and downstream in both seasons showed intermediary values. The smallest values of nuclear damage rate were found in REBIO and upstream in winter. In Comet Assay, it was verified through ANOVA, that the highest values of DNA fragmentation were found in downstream summer and the smallest values in REBIO summer and winter. These outcomes suggest that the REBIO are suffering an influence of substances that cause citotoxicity, while the sampled sites in downstream and upstream of Campo Mourão are affected probably for substances with genotoxic action.pt_BR
